The Orion™ platform from RareCyte® enables same-day 21-plex fluorescence tissue analysis

Multiplex tissue imaging has long been limited by fluorescence overlap, few simultaneous targets, and low throughput; this SITC 2020 poster shows the Orion platform resolving a 20-plex whole-slide panel from a single stain and a single scan, with results in hours rather than days on a system that images up to 21 channels.

  • A single 20-plex TissuePlex panel — a SYTOX Blue stain paired with antibodies — imaged in one whole-slide scan. The core panel covered Pan-CK, E-Cadherin, CD31, SMA, CD20, CD45, CD4, CD8A, FOXP3, PD-L1, PD-1, CD68, Ki67 and PCNA among its 20 markers, while the Orion platform itself is built to image up to 21 channels.
  • One standard staining round and one scan, with results in hours rather than days. A single-staining protocol and a single-scan acquisition on the Orion Instrument produced the whole-slide 20-plex dataset, which a pathologist then reviewed to verify staining specificity across the panel.
  • Each of the 20 Orion channels was checked against single-marker IHC on a serial section. Comparison with 20 single-plex IHC stains showed qualitatively similar localization, and on the same tonsil the panel separated three distinct PD-L1 expression levels by tissue region as well as non-overlapping CD4 and CD8 T-cell subsets, with FOXP3 confined to the nucleus in regulatory T-cells.
